چکیده

Most phages form plaques which, when prepared by the agar-layer technique (M. H. Adams, Bacteriophages, Interscience Publishers, Inc., New York, 1959), are sufficiently pronounced to permit their accurate enumeration under appropriate conditions of illumination. Other phages, however, are difficult to detect and enumerate because their plaques are small or excessively turbid, or both. Phage 83, employed for the genetic analysis Baldwin, J. Bacteriol. 82:875, 1961). After incubation of the assay plates for 8 hr at 37 C, the plaques were sufficiently developed to be scored (Fig. 1). The assay plates containing the fully developed plaques were then flooded with 10 ml of Trypticase Soy Broth (TSB, BBL) containing 0.1% 2,3,5 -triphenyltetrazolium chloride (TTC, Nutritional Biochemicals Corp., Cleveland, Ohio). After incubation at 37 C for 20 min, the broth was

Enhanced contrast of bacteriophage plaques in Salmonella with ferric ammonium citrate and sodium thiosulfate (FACST) and tetrazolium red (TZR).

Visualization of bacteriophage plaques may be enhanced by addition of ferric ammonium citrate and sodium thiosulfate (FACST) or 2,3,5-triphenyltetrazolium chloride (tetrazolium red, TZR) to the soft agar layer of a traditional bacteriophage plaque assay. Background color from these reagents improved contrast between clear plaques and turbid host lawns in trypticase soy agar (TSA) plates. Enhanc...

Improved Procedure for Screening Expression Libraries for Novel Autoantigens

The standard method for immunoscreening of a cDNA expression library is time-consuming becauseof the production of a large proportion of false positives during the first and second round of screening. This problem is more important when a sensitive chemiluminescence detection system is used. Due tothe high sensitivity of the detection system, there is a need to avoid false posi...

ISOLATION OF LYTIC BACTERIOPHAGE AB72P AGAINST MULTI-DRUG RESISTANT ACINETOBACTER BAUMANNII ISOLATES OBTAINED FROM BURN INFECTION

Background & Aims: Acinetobacter baumannii is a gram-negative pathogen that causes a wide range of hospital-acquired infections. Due to its intrinsic traits and its remarkable abilities to quickly acquire resistance genes, it has become resistant to most antimicrobial agents and a major problem for hospitals. In recent years, application of lytic bacteriophages has been considered to eradicate ...

Investigating the Effect of Lytic Bacteriophages on Pseudomonas Aeruginosa Isolates with Multidrug Resistance (MDR) in Tehran in 2021

Introduction: Pseudomonas aeruginosa bacterium has a high resistance to most anti-Pseudomonas drugs, which has caused concern all over the world. Therefore, the effect of lytic bacteriophages on Pseudomonas aeruginosa isolates with multi-drug resistance is investigated in this research. Methods and Materials: Different strains of Pseudomonas aeruginosa bacterium were identified and isolated fr...

In Vitro Biocontrol of Escherichia coli Through the Immobilization of its Specific Lytic Bacteriophage on Cellulose Acetate Biodegradable Film

Background and Aims: Bacteriophages are mandatory bacterial parasites that are harmless to human and animal, which are used by dipping or spraying in food as natural antimicrobial agents. The use of these methods leads to wasting or trapping of phage in food, but its immobilization on the polymer surface facilitates the contact of phage with the host cell at the food surface. Therefore, the aim...
